Can you assign one role to every collaborator at once?

Yes. Shopify's collaborator guidance for organizations states it plainly:

Shopify documents those bulk steps for users generally: in the Users list select the users, click Assign role, pick a role, Continue, then select stores and Done. For a collaborator, that last step is not how access arrives.

Does the bulk role hand over access to your stores?

No. A role sets what a person may do inside a store, not which stores they are in. Shopify puts the limit flatly: "You can't grant collaborators access to multiple stores."

Reaching a second store follows its own rule, and one account opens one store.

Access is built request by request: "Collaborator accounts are created per store based on the store access requests from the collaborators." Approve one person on two stores and you hold two accounts under one name. Roles are an admin-side setting, so POS access for a collaborator account is its own question.

Who can run it, and does it need Shopify Plus?

Shopify names who handles collaborator roles in an organization: the organization owner and the Organization administrator can edit and accept the auto-generated collaborator user role, or assign existing roles; the store owner, the Store user administrator and the Organization user administrator can only assign existing roles.

The nearest plan line covers something else: adding users and assigning store roles, open to every plan but Basic and Starter.

Organizations exist on both sides of the Plus line; the route differs. Shopify states that "By default, stores on the Shopify Plus plan have access to organization features and settings." An owner of two or more of their own stores, all sharing one billing currency other than INR, can group them instead — the documented route off Plus, since Plus stores use expansion stores rather than grouping.
